چکیده

In this study, possible paracrine factors in adipose tissue from lean and obese subjects were sought. Conditioned media were prepared by incubation in alpha minimum essential medium of adipocyte precursors derived from lean and massively obese subjects. Adipocyte-precursor-derived conditioned media from the obese stimulated replication of cultured rat perirenal adipocyte precursors by about fourfold over control. The effect of media conditioned by precursors derived from lean subjects was much less evident. The mitogenicity of conditioned media was abolished by trypsin, indicating the protein nature of the mitogenic factor(s). Sephacryl S-200 chromatography of adipocyte-precursor-derived conditioned media from obese subjects revealed one major active fraction with molecular masses in the range of 25,000-40,000. Our results demonstrate that adipocyte precursors derived from massively obese subjects release factors mitogenic on cultured rat adipocyte precursors. These principles may act as paracrine factors contributing to the development of the adipocyte hyperplasia characteristic of massive obesity.

17-beta-estradiol increases mitogenic activity of medium from cultured preadipocytes of massively obese persons.

Having reported that omental preadipocytes from massively obese persons release into the culture medium proteins mitogenic for preadipocytes, this study aimed to determine whether estrogens contribute to the production of these factors. Sub-cultured omental preadipocytes from 13 massively obese women were grown in the presence or absence of 17-beta-estradiol, and during the last 24 h the condit...

Stress and Atherogenesis: Smooth Muscle Cell Mitogenic Activity and other Biochemical Changes Associated with Sera of \"Stressed\" Subjects

The proliferation of smooth muscle cells in the arterial wall (VSMC) is considered to play a key role in the development of atherosclerosis. To investigate the possible contribution of "stress" (experimentally-induced) to this process, blood from healthy volunteers, ages 21 to 65, screened to exclude major risk factors for coronary heart disease, was assayed for mitogenic activity after the sub...

Fat depot origin affects adipogenesis in primary cultured and cloned human preadipocytes.

Fat distribution varies among individuals with similar body fat content. Innate differences in adipose cell characteristics may contribute because lipid accumulation and lipogenic enzyme activities vary among preadipocytes cultured from different fat depots. We determined expression of the adipogenic transcription factors peroxisome proliferator activated receptor-gamma (PPAR-gamma) and CCAAT/e...

Identification of Depot-Specific Human Fat Cell Progenitors through Distinct Expression Profiles and Developmental Gene Patterns Running title: Preadipocytes from Different Depots Are Distinct

Anatomically separate fat depots differ in size, function, and contribution to pathological states such as the metabolic syndrome. We isolated preadipocytes from different human fat depots to determine if the basis for this variation is partly attributable to differences in inherent properties of fat cell progenitors. We found that genome-wide expression profiles of primary preadipocytes cultur...
